RBP Target Engagement (RBP-TE™) is used when mechanistic hypotheses or RNA-binding protein targets are already defined. By directly measuring compound-induced changes in endogenous RNA–protein interactions in cells, the assay provides focused, quantitative evidence of cellular target engagement.
This service lets you establish quantitative RBP-TE™ assays in your selected cellular matrix, enabling RNA–protein interaction–coupled target engagement measurements under physiologically relevant conditions, without tags, overexpression, or reporter systems. The assay can be used to generate IC₅₀ values based on in-cell target engagements, supporting hit-to-lead decisions by differentiating on-target engagement from off-target effects and identifying cases where downstream phenotypic responses are not supported by direct target engagement.
As a result, RBP-TE™ is well-suited for validating mechanistic hypotheses and confirming that compounds engage the intended RNA-binding protein(s) in their native cellular context. The resulting cellular target engagement IC₅₀ values and dose–response diagnostics support confident hit confirmation, engagement-informed hit-to-lead progression, and early lead optimization by enabling quantitative SAR interpretation, compound ranking, and lead series prioritization.
